新宿上空に突如、妖邪界の阿羅醐城が出現。妖邪帝王阿羅醐が妖邪四天王と呼ばれる４人の戦士、朱天、螺呪羅、那唖挫、悪奴弥守と妖邪兵を指揮して人間界の支配に乗り出す。それに対抗するのは、ヨロイギアと呼ばれるアーマーを装着することで鎧武者の戦士へと変身を遂げる５人の少年たち。烈火のリョウこと真田遼、水滸のシンこと毛利伸、天空のトウマこと羽柴当麻、金剛のシュウこと秀麗黄、光輪のセイジこと伊達征士。サムライトルーパーと呼ばれる彼らは、戦国の世から妖邪と戦ってきた迦雄須と名乗る雲水の助けを借りて阿羅醐と戦っていく。
